There is no “spiritual” meaning of 666. It is the numerical equivalent of the spelling of Nero Caesar.
666=18.

The “mark of the Beast” seems to be some sort of passport in the form of a tattoo for those who want to trade within the Roman Empire, or maybe some kind of proof that someone is part of the Empire and entitled to trade within it. Whether it is some kind of branding or something else may be debateable.
